A High Parameter Flow Cytometric Assay to Identify Human Myeloid Derived Suppressive Cells

چکیده
Abstract The concept of myeloid derived suppressive cells (MDSCs) has emerged in recent years as a group that pocess potent immunosuppressive activity. These expand under pathologic conditions and suppress the immune functions T cells, dendritic macrophages, natural killer cells. They have been shown to be involved pathological states cancer, autoimmune diseases, infections, such COVID-19. MDSCs are subset into PMN (polymorphonuclear)- M (monocytic)-MDSCs. Phenotypic markers elucidated years. Here Cytek® developed 14-color, 16-marker, single-tube flow cytometric assay analyzed MDSCs. panel includes exclude lymphoid lineage identify PMN-MDSC M-MDSC populations, CD15, CD14, CD16, CD66b, CD11b HLA-DR. In addition, PMN- specific expression Lox-1, CXCR-1, DR5 also included. Fresh whole blood samples were stained with erythrocyte lyse wash method on Aurora or 3-laser Northern Light TMinstrument. performance was evaluated collected EDTA, Heparin, ACD, Cytochex tubes well peripheral mononuclear cell (PBMC) samples. Assay precision assessed by testing 12 donor triplicates three consecutive days. Targeted populations identified PBMC good resolution %CV less than 25% for all interests. This 14-color 16-marker preoptimized can used clinical research drug discovery infections other inflammatory diseases.

A rapid and sensitive intracellular flow cytometric assay to identify Theileria parva infection within target cells.
Theileria parva is an intracellular protozoan parasite transmitted by ticks that causes a fatal lymphoproliferative disease of cattle known as East Coast Fever. Vaccination against the disease currently relies on inoculation of the infective sporozoite stage of the parasite and simultaneous treatment with long-acting formulations of oxytetracycline. Sporozoites are maintained as frozen stabilat...
متن کاملComparison of Two Flow Cytometric Methods for Detection of Human Invariant Natural Killer T Cells (iNKT)
Background: Invariant natural killer cells (iNKT) are an important immunoregulatory T cell subset. Currently several flow cytometry-based approaches exist for the identifi-cation of iNKT cells, which rely on using the 6B11 monoclonal antibody or a combina-tion of anti-Vα24 and anti-Vβ11 antibodies. Objective: The aim of this study was to compare the ability of two flow cytometry-based methods f...
متن کاملMicroRNA-155 regulates tumor myeloid-derived suppressive cells
MicroRNA is small non-coding RNA and can lead to translational repression or target degradation by base-pairing with complementary sequences of mRNA molecules. MicroRNA-155 (miR-155), one of the most studied microRNA, is the first one to be reported as oncogenic [1]. miR-155 is over expressed in a long list of both hematological and solid tumors and is of paramount importance in cancer diagnosi...
متن کاملEstablishment of lal-/- Myeloid Lineage Cell Line That Resembles Myeloid-Derived Suppressive Cells
Myeloid-derived suppressor cells (MDSCs) in mouse are inflammatory cells that play critical roles in promoting cancer growth and metastasis by directly stimulating cancer cell proliferation and suppressing immune surveillance. In order to facilitate characterization of biochemical and cellular mechanisms of MDSCs, it is urgent to establish an "MDSC-like" cell line. By cross breeding of immortom...
